Description

Seeking an undergraduate of junior standing to work on a project constructing artificial rosettes from a bacterial cellulose synthase. The person will work under the supervision of Research Associate Professor Sung Hyun Cho and Graduate Student Lynnicia Massenberg. With their guidance, the student will clone DNA encoding oligomerizing helicees adjacent to the coding region of a gene encoding a bacterial cellulose synthase. Once prepared, the genes will be expressed in E. coli cells, the recombinant proteins purified by column chromatography and tested for the ability to make cellulose in the forms of single beta-glucan chains (a chemical assay) and cellulose microfibrils (a microscopic assay). Depending on the observed functions, purified protein will be examined by structural methods including Negative Stain TEM, cryo-tomography, and single particle cryo-TEM. It is required that the student be able to commit to work through July of 2020. The position can start immediately. This is a valuable opportunity to learn molecular biology and microscopy techniques while earning co-authorship on a future manuscript describing current research. Qualified applicants with background in biochemistry, microbiology, biology, chemistry, chemical or biochemical engineering will be considered.
